Smaller Snacking is Smart Snacking: New study shows 'just a bite' will satisfy

Jan. 29, 2013 ? How much chocolate would you need to eat to be satisfied? Less than half as much as you think, according to this recently published Cornell University snacking study. Using chocolate chips, apple pie, and potato chips, researchers Ellen van Kleef, Mitsuru Shimizu, and Brian Wansink designed a study to determine if people who were given smaller portions of snack foods would feel hungrier or satisfied fifteen minutes after eating.

Two groups with different portion sizes were tested. The larger portion size group was given 100g of chocolate, 200g of apple pie, and 80g of potato chips, all slightly larger than the recommended portion sizes. This equaled 1370 calories in snack foods. The other group was given 10g, 40g, and 10g of these same foods respectively, for a total of 195 calories. The two groups were given as much time to eat as needed, and were asked to fill out surveys to rate the liking, familiarity, and boredom with the food. They were also asked to rate their hunger and craving before the food was presented and fifteen minutes after the taste tests ended.

The results remarkably showed that smaller portion sizes are capable of providing similar feelings of satisfaction as larger ones. Those given larger portions consumed 77% more food, amounting to 103 calories more, but they did not feel any appetite enhancing or stronger feelings of satiety than the group with the smaller portions. Overall these findings reflect the importance of portion size. While larger portions result in increased food intake, smaller portions may make you feel equally satisfied. The smaller portions can lead to a decline in hunger and desire that would help people limit their food intake. So, next time you are craving a snack food, remember that you can feel similarly satisfied with one handful as you would with two!

Fragile economy, other global woes dominated Davos

DAVOS, Switzerland (AP) -- The fragile state of the world economy, along with the relentless turmoil in Syria and the rocky fallout from the Arab Spring, dominated discussions during this year's annual gathering of the global elite at Davos, leaving many participants uneasy about what lies ahead as they left for home Sunday.

Even broad agreement that there are some positive signs on the economic front, at least in emerging markets, was coupled with a warning from the head of the International Monetary Fund. "Do not relax," Christine Lagarde said. There's still a "risk of relapse."

More than 2,500 of the best and brightest in business, government, academia and civic life gathered for the five-day World Economic Forum at this Alpine resort. But much of the overt glitz and glamor that is a usual feature was toned down or absent this year, a decision founder Klaus Schwab said reflected the serious issues facing the world.

Political and economic issues vie for top billing each year at Davos, and this time, the economy had the edge, with a special focus on how to promote economic growth and jobs, especially for the youth among the world's 220 million jobless.

The IMF said that China, Africa, and other emerging markets could see significant growth, but Japan, eurozone nations and the U.S. are likely to struggle with negative to low growth. Ahead of the 43rd forum, the IMF downgraded its forecast for global economic growth this year by one-tenth of a percentage point to 3.5 percent.

While the U.S. avoided the so-called "fiscal cliff" of automatic tax increases and spending cuts, and fears have abated that the euro currency union will break up, there is growing concern that governments may ease up on measures to improve growth and reduce debt that the IMF and many other institutions are calling for.

IMF chief Lagarde said the "very fragile and timid recovery" depends on leaders in the 17-nation eurozone, the United States and Japan making "the right decisions." The eurozone in particular "is fragile because it is prone to political crisis" and slow decision-making, she said.

Davos participants' uneasiness about the world economy was matched by growing concern over the political turmoil in the Arab world, terrorism in North Africa, a spate of natural disasters that have highlighted the failure to tackle climate change, and the growing inequality between the world's "haves" and "have nots."

"Two years ago, gloom around the stalled economic recovery was leavened by euphoria at the outbreak of the Arab spring," Kenneth Roth, the executive director of Human Rights Watch, told The Associated Press at Saturday night's low-key final reception. "This year, relief at the improved economic outlook is tempered by despair at the unimpeded slaughter in Syria, uncertainty about the outlook in Egypt, and frustration over the Arab monarchies' resistance to reform."

The Arab Spring uprisings have ousted dictators in Tunisia, Yemen, Libya and Egypt over the past two years. But now Islamists and liberals are wrangling over power, with Islamists mainly gaining the upper hand. Democracy is far from certain, and economic woes have left hundreds of thousands of young people jobless and frustrated that their "revolutions" haven't produced any dividends.

Former Arab League Secretary-General Amr Moussa, a losing candidate in Egypt's presidential election last year, said there have been achievements, but warned that democracy isn't only about casting a vote.

"It is the respect of human rights, for rights of women, separation of powers, independence of the judiciary. This meaning of democracy we have not yet achieved," Moussa said.

Elsewhere in the Middle East, Israeli-Palestinian peace talks remain stalled, Arab monarchs remain entrenched, and the death toll from the escalating civil war in Syria has topped 60,000 with no end in sight.

Jordan's King Abdullah II, whose country is hosting almost 300,000 Syrian refugees, predicted that Syrian President Bashar Assad's regime will last at least another six months. He called for a transition plan involving all Syrians and the Syrian army.

He also urged stepped up international support to end the Syrian crisis, saying, "The weakest refugees are struggling now just to survive this year's harsh winter."

Abdullah told the forum that "unprecedented threats to regional and global stability and security" need international action now, not the "wait and see" response by some countries ? which he did not identify ? especially in helping governments emerge politically and financially from the Arab uprisings.

The king, considered one of the region's moderate leaders, also warned Israel to stop playing the "waiting game," and said President Barack Obama's second term offered the last opportunity to create two states ? Palestine and Israel ? that can live side-by-side in peace.

Angel Gurria, secretary-general of the Paris-based Organization for Economic Cooperation and Development, said the focus on resolving the world's economic crisis has distracted leaders from many other important issues, including education, the social consequences of unemployment and promoting ways to deal with climate change.

Nonetheless, Gurria said, the world should be "very worried" because there aren't many "tools" left to fix the economy if things get worse.

Trevor Manuel, South Africa's National Planning Commission minister, told AP that the key message from Davos for him was a positive one ? that "many of the decisions that have been taken bring us closer to where we need to be." He warned that "a sense of an all-pervasive gloom ... frequently becomes a self-fulfilling prophecy."

(AP) ? Possible No. 1 ranking? Yeah, that's nice, Michigan's Trey Burke said Sunday outside the locker room at Illinois' Assembly Hall.

But after the win that should give the No. 2 Wolverines their first top ranking since the 1992-93 season, the sophomore guard wanted to talk defense and what a team does when one of its big, defensive rocks goes down.

If you're Michigan (19-1, 6-1 Big Ten), you plug in a couple of other big guys. And you win ? in this case 74-60 over the Illini, most of it without 6-foot-8, 250-pound Jordan Morgan. The starting forward limped to the locker room less than two minutes into the game with a sprained right ankle and didn't come back.

"Coach (John Beilein) calls him the minister of defense," Burke said, crediting the Mitch McGary, John Horford and Max Bielfeldt, the three big men who picked up the slack.

"I say this every game," Burke added, "but it just starts with defense. I think that started in the first half."

And that No. 1 ranking, which will likely be Michigan's after Duke's lopsided loss to Miami earlier in the week opened the door when the AP poll comes out Monday, doesn't mean much yet, Beilein was quick to remind.

"You compete for a Big Ten championship, then you go on you compete for a national championship," he said. "That's the number one you want down the line."

Illinois (15-6, 2-5) and first-year coach John Groce, whose team has been doomed by cold shooting since mid-December and shot 37.1 percent Sunday, could do little but marvel at the Wolverines.

"I give Michigan a lot of credit," Groce said. "You make one mistake and they exploit it like that."

Burke scored 19 to lead Michigan. Nick Stauskas scored 14 points and Glenn Robinson III and Tim Hardaway Jr. added 12 each.

Brandon Paul led Illinois with 15 points but had five of the Illini's 15 costly turnovers.

Beilein said he didn't yet know the severity of Morgan's sprained right ankle but said he couldn't have returned to the game even if baldy needed.

The Wolverines hardly missed him ? they seemed to have an answer for everything Illinois offered.

The Illini fought back to within seven points with just over nine minutes to play when a Richardson steal set up Joseph Bertrand for a soft jump shot that floated into the bucket.

The wave of noise that rose from the crowd trying to will the Illini back into the game didn't last long.

First, with 8:40 to play, Mitch McGary pulled down the rebound off a miss by Burke and dropped the ball into the bucket.

Then Burke scooped the ball up off a Paul turnover at the other end and, with a dunk, put the Wolverines back up by 11 at 59-48. With 8:21 to play and shooting just 37.1 percent on the night, Illinois couldn't find a way back.

Morgan appeared to roll his right ankle as he came down under the Wolverine basket, but Michigan lost little if anything inside without the 6-foot-8, 250-pound forward.

McGary, Bielfeldt ? a redshirt freshman with strong ties to Illinois ? and Horford picked up most of Morgan's minutes, and his slack.

McGary, a 6-10, 250-pound forward, hasn't started a game this season but averages 16 minutes a night anyway. Bielfeldt, though, plays less than six minutes a night, and was all nerves in his first minutes on the court. The 6-7, 245-pound forward badly missed his first free throw, at least a foot right of the basket, and the crowd, well aware that the athletic administration building on the Illinois campus bears his big-donor family's name, let him have it.

So did his teammates, Beilein said.

"It was not a great initial debut with the air ball. That's one of the first things we made fun of," the coach said. "All his teammates were on him in the locker room."

But with another chance at the line minutes later, Bielfeldt sank both shots, finishing with four points in six minutes.

Horford had seven points and five boards in 17 minutes while McGary had six points and eight rebounds.

"It gave three guys the opportunity to play that (don't) get as much as they like, Beilein said of Morgan's injury. "I'm really impressed with the big guys and how they stepped up there."

Illinois' big-man combination of Nanna Egwu, Sam McLaurin and Tyler Griffey wasn't much of a match for them. The Wolverines scored 42 points in the paint.

Groce said his big men weren't bad, just not as good they'd like to be, especially McLaurin, the 6-8 grad-student transfer he called his team's defensive Kevin Garnett

"I really have a high regard for Sam's intelligence and ability to execute our defense, and I think we'll look at the film and he wasn't on his Ps and Qs like normal," said Groce, who last faced Michigan as the coach at Ohio, leading the Bobcats to a second-round NCAA Tournament upset last spring.

Michigan took the lead for good with 15:21 to play in the first half when Burke hit a jump shot to go up 8-7. With a 9-0 run that ended only with a 3-pointer by Paul with 10:52 left in the half, the Wolverines were up 15-10.

Iraq: 2 soldiers killed near rally shooting site

FALLUJAH, Iraq (AP) ? Gunmen killed two Iraqi soldiers and abducted three others in Fallujah on Saturday as hundreds of mourners gathered in the restive western city for the funeral of protesters killed during a shooting by army troops a day earlier, according to officials.

The attacks and kidnappings appeared to be in retaliation for the deaths of protesters in clashes Friday, and are likely to further strain tensions between Iraq's minority Sunnis and the Shiite-led central government.

Also on Saturday, Iraqi lawmakers said parliament has approved a law that would limit the terms of the prime minister, president and parliament speaker to a maximum of two terms. The measure, which must still be approved by Iraq's president, could pose a challenge to Prime Minister Nouri al-Maliki's plans to seek the post again in 2014.

Fallujah police Maj. Rasheed al-Adeeli said one of the soldiers killed was hit by a sniper on the outskirts of the city. Another was shot dead when gunmen attacked a military post where soldiers were packing up their equipment on the northern edge of the city.

Gunmen ambushed the car of three off-duty soldiers on the outskirts of Fallujah and kidnapped them, according to state television and a provincial police official reached by The Associated Press. The official, who spoke on condition of anonymity because he was not authorized to release the information, said authorities have launched a manhunt for the missing men.

Mourners hoisted caskets and waved Iraqi flags while shouting "Allahu Akbar!" or God is great. The funeral procession took place in central Fallujah, not far away from the city's cemetery.

Iraqi soldiers opened fire Friday on stone-throwing demonstrators near Fallujah, killing at least five, according to local authorities. Two other soldiers were killed Friday in apparent payback for the protesters' deaths.

Sunnis angry over perceived second-class treatment and what they see as unfair policies targeting their sect began demonstrating last month in Anbar. The protests have since spread to other predominantly Sunni areas.

The protesters are demanding the release of detainees and the cancellation of a tough counterterrorism law and other policies they believe overwhelmingly target Sunnis. Many link their cause with the broader Arab Spring and are calling for the downfall of the government altogether.

International rights group Amnesty International pressed the Iraqi government to immediately investigate the protesters' deaths and make its findings public.

"Anyone found responsible for abuses ? including anyone found to have used excessive force against protestors ? must be brought to justice," said Ann Harrison, Amnesty's deputy program director for the region. She also urged authorities to ensure security forces are properly trained and equipped to deal with demonstrators in a way that respects human rights.

Iraqi state television aired a statement from the Defense Ministry on Friday evening saying it would investigate what happened in Fallujah. Repeated efforts to reach ministry officials directly were unsuccessful.

The United Nations envoy to Iraq, Martin Kobler, said the U.N. is increasingly concerned about the rising level of tension within the country. He said he would welcome a decision by the defense ministry to investigate the incident.

"We urge the government to exert self-restraint in law and order, and respect human rights," he said in an interview. "Our message to the demonstrators is to exercise their right to demonstrate peacefully."

Kobler also called on political leaders across Iraq to "sit together in a constructive dialogue."

Muhammed al-Khalidi, a Sunni lawmaker, said that 170 lawmakers voted in favor of the law limiting the terms on the prime minister and other top officials. He said lawmakers from al-Maliki's State of Law bloc boycotted the session.

Mushriq Naji, a lawmaker from the bloc headed by firebrand cleric Moqtada al-Sadr, confirmed the vote. He said the measure is needed "to prevent the return of the dictatorship to Iraq."

"This law ensures that no person can stay in power for an unlimited time and that democracy will continue in this country," Naji said.

Putting the law into force could prove challenging, however.

Iraq's president must sign off on the legislation. The current president, Jalal Talabani, is incapacitated following a stroke and is being treated in a German hospital, and it is unlikely he would be able to sign the law into force.

Legal expert Tariq Harb downplayed the significance of the vote. Even if it were to be enacted, the supreme court could rule that it contradicts the constitution and is therefore invalid, he said.

"This vote could be part of a political game because the Iraqi constitution does not limit the terms for the prime minister and the parliament speaker," he said. "Also, the same parliament could convene at any time in the future and decide to abolish the new law if the political map changes."

Numark iDJ Live II DJ controller hands-on (video)

Numark iDJ Live II DJ controller hands-on

If there's one company that comprehensively caters to the entry-level and intermediate DJ, there's a good chance it's Numark. While the firm has released a whole bunch of stuff at NAMM this year, perhaps its most mainstream offering is the iDJ Live II. If you remember the original, it was a lightweight controller (both physically, and metaphorically) designed for use with Djay on iOS. This time around the little fella gets a redesign, and a few functionality tweaks. Most notable, is that while the first iteration only gave you the option of iOS connectivity -- via the 30-pin adaptor -- this time you have the choice of Lightning, 30-pin, or straight up USB. Thus, you can now bring the iDJ Live II out to work with your Mac or PC, too. Once again, it's built with Algoriddim's Djay in mind, be it the desktop or iOS version, and we got our hands on it to take it for a spin. Head past the break for the lowdown.

First things first. This is very much a fun, consumer product. It would be unfair to call it an accessory, but it's definitely meant for casual use at parties etc. The unit itself is very light, made entirely of plastic, and feels very different to the heavier DJ controllers you might be used to. The iDJ Live II, however, does look much better than the first iteration, with the slightly squared design and the lower-profile platters at least making it look a little less toy-like than before. The plastic finish still feels the same, as does the action and resistance of the platters, knobs and faders (which is serviceable). The controls available give you everything you need to perform all the mixing basics, with high and low-frequency EQ, channel volume control, a crossfader, and even rudimentary scratching (though it really is more of a novelty). If you've ever used Djay for any amount of time though, you'll know that this is plenty enough to get a set going, and having the physical controller there just makes it all that more enjoyable / easy. While purists and old hands might see products such as these as over simplified, the ease of use, and broad appeal makes them ideal for the curious, and that can only be a good thing. Sadly there is no word on price or availability at this time, but we'd be surprised if it wasn't comfortably under $100 when it does finally make its way onto shelves. Check the video below for the hardware tour.

What's the deal at Sundance? Market gains steam with more buyers

NEW YORK (TheWrap.com) - Movies starring Joseph Gordon-Levitt and Steve Carell fetched millions this year at the Sundance Film Festival as sellers adjust to a market that is more diffuse than in years past but is also steadily gaining steam.

Major distributors like Relativity and Paramount surprised many by acquiring or bidding on films, but the biggest acquisitions so far have been for movies with A-list stars in the cast. Fox Searchlight paid $9.5 million for "They Way, Way Back," which features Carell and Toni Collette. Relativity paid $4 million and promised $25 million more in marketing for "Don Jon's Addiction," which stars Gordon-Levitt, Scarlett Johansson and Julianne Moore.

Acquisitions picked up this week after a sleepy weekend, with a half-dozen films closing deals and Sony Pictures Classics closed a deal late Tuesday night for "Kill Your Darlings," starring Daniel Radcliffe as poet Allen Ginsberg.

This year has mirrored the trend of the recent past, with deal-making more cautious and less impulsive, demonstrating that Sundance movies now attract all kinds of buyers, not only theatrical distributors.

"The last couple years, deals have been closing on Sunday and Monday instead of right away," David Glasser, chief operating officer of the Weinstein Company, told TheWrap. "We sit with the film, run the models and see how far we're willing to go."

Halfway into the festival, no film has become the talk of the town, as "Beasts of the Southern Wild" and "The Surrogate" were last year. Nor has any company taken the market by storm.

The Weinstein Company often fills that role, as in 2011 when it paid about $13 million for "Our Idiot Brother" and "The Details." And at Toronto in 2012, Lionsgate and Roadside Attractions took "Thanks for Sharing," "Imogene" and "Much Ado About Nothing."

At Sundance this year, Weinstein took "Fruitvale," and Roadside remains on the sidelines. Newer distributors, such as The Weinstein Company's multi-platform label RADiUS and A24 took popular titles "Concussion," "Lovelace" and "The Spectacular Now." Exclusive Releasing acquired the divisive "Two Mothers," its first title since its September launch.

"It's a phenomenon of films finding the right distribution rather than just going up for auction," said John Sloss, founder of Cinetic Media and a veteran seller. "Do I miss the days when $4 million deals were the norm? It made my deals easier. But if you have to wait, you collect on the back end."

Sloss' slate typifies the new market. His "Toy's House," sold late Tuesday to CBS Films. Other titles, such as "Before Midnight" and "Blue Caprice," have received warm receptions but are still up for sale. A large number of buyers are in pursuit, reflecting an infusion of new companies into the marketplace.

Sloss dubbed the "Don Jon" and "Way, Way Back" deals "an anomaly," but Glasser pointed to Sony's $4 million acquisition of " Austenland" and his own "Fruitvale" deal as signs of a strong year.

"There's only so much any of us can take on," Glasser said. "You pick those one or two movies you really go after and this year it ended up being spread among all of us. It's very interesting how the VOD market, especially our division, has come into play."

RADiUS is one of several companies that offers new-release options to filmmakers, promising that video on demand and digital sales can be as lucrative as a theatrical release or big upfront money.

"We have to look as much at ancillary revenue streams and back end money as the advance," Sloss said. "It feels like there's a lot more thoughtfulness from sellers and buyers."

Documentaries are the one category of film that are doing brisk business, from the killer whale documentary "Blackfish" to the immigrant tale "Who is Dayani Cristal?"

With the festival halfway over, the question is when the second flurry of deals for narrative films will begin. Lynn Shelton's "Touchy Feely," horror anthology "S-VHS" and Richard Linklater's "Before Midnight" are among the many titles expected to go soon. If the market so far has been any indication, expect multiple bidders and smaller advances - but plenty of deals.

A $17.8 million test project will send to the International Space Station an inflatable room that can be compressed into a 7-foot tube for delivery, officials said Wednesday in a news conference at North Las Vegas-based Bigelow Aerospace.

If the module proves durable during two years at the space station, it could open the door to habitats on the moon and missions to Mars, NASA engineer Glen Miller said.

The agency chose Bigelow for the contract because it was the only company working on inflatable technology, said NASA Deputy Administrator Lori Garver.

Founder and President Robert Bigelow, who made his fortune in the hotel industry before getting into the space business in 1999, framed the gambit as an out-of-this-world real estate venture. He hopes to sell his spare tire habitats to scientific companies and wealthy adventurers looking for space hotels.

NASA is expected to install the 13-foot, blimp-like module in a space station port by 2015. Bigelow plans to begin selling stand-alone space homes the next year.

The new technology provides three times as much room as the existing aluminum models, and is also easier and less costly to build, Miller said.

Artist renderings of the module resemble a tinfoil clown nose grafted onto the main station. It is hardly big enough to be called a room. Miller described it as a large closet with padded white walls and gear and gizmos strung from two central beams.

Garver said Wednesday that sending a small inflatable tube into space will be dramatically cheaper than launching a full-sized module.

"Let's face it; the most expensive aspect of taking things in space is the launch," she said. "So the magnitude of importance of this for NASA really can't be overstated."

The partnership is another step toward outsourcing for NASA, which no longer enjoys the budget and public profile of its heyday. The agency has handed off rocket-building to private companies, retired it space shuttles in 2011 and now relies on Russian spaceships to transport American astronauts to and from the space station.

Astronauts will test the ability of the bladder, known as the Bigelow Expandable Activity Module, or BEAM, to withstand heat, radiation, debris and other assaults. Some adventurous scientists might also try sleeping in the spare room, which is the first piece of private real estate to be blasted into space, Garver said.

Bigelow said the NASA brand will enable him to begin selling Kevlar habitats several times the size of the test module.

"This year is probably going to be our kickoff year for talking to customers," he said. "We have to show that we can execute what we're talking about."

Bigelow, who launched a small prototype of the module in 2006 after licensing the patent from NASA, will rely on Boeing Co. and Southern California rocket developer Space Exploration Technologies to provide transportation.

A 60-day stay will cost $25 million, which doesn't include the $27.5 million it costs to get there and back.

Bigelow predicted that the primary customers will be upwardly mobile countries including Brazil, Singapore and the United Arab Emirates that "have a difficult time getting their astronauts into orbit" and could use a private space station to barter and build up prestige.

The biggest technological challenge will be transporting the collapsed module through the sub-zero temperatures of space without tearing or cracking any part of it, Miller said.

When it arrives at the space station in 2015, scientists will blow it up and let it sit for a few days to test for leaks. If it does not hold as promised, NASA will take back a portion of the already bargain basement price it paid Bigelow.

Standing beside scale models of research stations on Mars and the moon, Miller said the project will encourage commercial ventures to follow the path NASA blazes into space.

He added that it could also help achieve the holy grail of space exploration: missions that send astronauts out of orbit for more than a year.

"The only way to do that is to expand it out and voila you have living space for three people to go to Mars," he said. "You can get three times the volume of a metallic can, and you can go up in the same ferry."

Retrovirus in the human genome is active in pluripotent stem cells

Thursday, January 24, 2013

A retrovirus called HERV-H, which inserted itself into the human genome millions of years ago, may play an important role in pluripotent stem cells, according to a new study published in the journal Retrovirology by scientists at UMass Medical School. Pluripotent stem cells are capable of generating all tissue types, including blood cells, brain cells and heart cells. The discovery, which may help explain how these cells maintain a state of pluripotency and are able to differentiate into many types of cells, could have profound implications for therapies that would use pluripotent stem cells to treat a range of human diseases.

"What we've observed is that a group of endogenous retroviruses called HERV-H is extremely busy in human embryonic stem cells," said Jeremy Luban, MD, the David L. Freelander Memorial Professor in HIV/AIDS Research, professor of molecular medicine and lead author of the study. "In fact, HERV-H is one of the most abundantly expressed genes in pluripotent stem cells and it isn't found in any other cell types."

In the study, Dr. Luban and colleagues describe how RNA from the HERV-H sequence makes up as much as 2 percent of the total RNA found in pluripotent stem cells. The HERV-H sequence is controlled by the same factors that are used to reprogram skin cells into induced pluripotent stem (iPS) cells, a discovery that garnered the 2012 Nobel Prize in Physiology or Medicine. "In other words, HERV-H is a new marker for pluripotency in humans that has the potential to aid in the development of iPS cells and transform current stem cell technology," said Luban.

When a retrovirus infects a cell, it inserts its own genes into the chromosomal DNA of the host cell. As a result, the host cell treats the viral genome as part of its own DNA sequence and begins making the proteins required to assemble new copies of the virus. And because the retrovirus is now part of the host cell's genome, when the cell divides, the virus is inherited by all daughter cells.

In rare cases, it's believed that retroviruses can infect human sperm or egg cells. If this happens, and if the resulting embryo survives, the retrovirus can become a permanent part of the human genome, and be passed down from generation to generation. Scientists estimate that as much as 8 percent of the human genome may be comprised of extinct retroviruses left over from infections that occurred millions of years ago. Yet these sequences of fossilized retrovirus were thought to have no discernible functional value.

"The human genome is filled with retrovirus DNA thought to be no more than fossilized junk," said Luban. "Increasingly, there are indications that these sequences might not be junk. They might play a role in gene expression after all."

An expert in HIV and other retroviruses, Luban and his colleagues were seeking to understand if there was a rationale behind where, in the expansive human genome, retroviruses inserted themselves. Knowing where along the chromosomal DNA retroviruses might attack could potentially lead to the development of drugs that protect against infection; better gene therapy treatments; or novel biomarkers that would predict where a retrovirus would insert itself in the genome, said Luban.

Turning these same techniques on the retrovirus sequences already in the human genome, they discovered a sequence, HERV-H, that appeared to be active. "The sequences weren't making proteins because they had been so disrupted over millions of years, but they were making these long, noncoding RNAs," said Luban.

Specifically, the HERV-H sequence was making abundant amounts of RNA in human embryonic stem cells?and only stem cells. In total, there are more than 1,000 HERV-H retrovirus genomes scattered throughout the human genome. The Luban lab also found high levels of HERV-H RNA in some iPS cells. Other iPS cells, perhaps those lines that were not sufficiently reprogrammed to pluripotency, had lower levels of the HERV-H RNA, another indication that HERV-H may be an important marker for pluripotency.

Interestingly, the HERV-H genes that were expressed in human pluripotent stem cells are only found in the human and chimpanzee genomes, indicating that HERV-H infected a relatively recent ancestor to humans, said Luban.

"Once upon a time HERV-H was an invader to our genome and perhaps caused diseases like AIDS or cancer," said Luban. "Now it seems that a kind of d?tente has been reached. Not only that, but this ancient invader may one day be exploited by clinicians to cure people of a wide range of diseases using stem cell therapies."

Luban and colleagues will next try to determine the specific mechanisms by which HERV-H contributes to pluripotency.

U.S. homeland chief: cyber 9/11 could happen "imminently"

WASHINGTON (Reuters) - Homeland Security Secretary Janet Napolitano warned on Thursday that a major cyber attack is a looming threat and could have the same sort of impact as last year's Superstorm Sandy, which knocked out electricity in a large swathe of the Northeast.

Napolitano said a "cyber 9/11" could happen "imminently" and that critical infrastructure - including water, electricity and gas - was very vulnerable to such a strike.

"We shouldn't wait until there is a 9/11 in the cyber world. There are things we can and should be doing right now that, if not prevent, would mitigate the extent of damage," said Napolitano, speaking at the Wilson Center think tank in Washington and referring to the September 11, 2001, attacks.

Napolitano runs the sprawling Homeland Security Department that was created 10 years ago in the aftermath of September 11 and charged with preventing another such event.

She urged Congress to pass legislation governing cyber security so the government could share information with the private sector to prevent an attack on infrastructure, much of which is privately owned.

A cyber security bill failed in Congress last year after business and privacy groups opposed it. The measure would have increased information-sharing between private companies and U.S. intelligence agencies and established voluntary standards for businesses that control power grids or water treatment plants.

Business groups said the legislation was government overreach. Privacy groups feared it might lead to Internet eavesdropping.

New cyber legislation is being considered, but it is unclear whether it will get through the gridlocked Congress.

President Barack Obama is expected to soon issue an executive order that would set up a voluntary system to help protect some critical infrastructure and offer incentives to companies that participate.

But without a new law, companies cannot be granted any kind of legal immunity for sharing information with the government and within the industry about potential threats.

Officials have pointed to recent hacking attacks on U.S. banks as a sign that the cyber threat is real and growing.

"The clarion call is here and we need to be dealing with this very urgently," said Napolitano. "Attacks are coming all the time. They are coming from different sources, they take different forms. But they are increasing in seriousness and sophistication."

Obama adviser optimistic on immigration overhaul

WASHINGTON (AP) ? A top White House adviser said Sunday the stars seem aligned for Congress to pass a comprehensive immigration overhaul this year, but he sounded less confident about prospects for toughening the nation's gun laws.

White House Senior Adviser David Plouffe made the rounds on Sunday talk shows, outlining the president's agenda for the months ahead. He said past presidents have been able to make significant progress during their second terms, noting that President Ronald Reagan pushed through more tax cuts and that President Bill Clinton helped transform budget deficits into budget surpluses.

He said Obama's focus will be on improving the economy, saying the president believes the best way to do that is to invest in education and manufacturing while also seeking what he called "balanced deficit reduction."

Republicans agreed to let tax cuts expire this year for those workers whose incomes exceed $400,000 a year, but Plouffe said that future negotiations on reducing the deficit will have to include more tax revenue as well as spending cuts and changes to entitlement programs.

"We've dealt with the tax rate issue. Now it's about loopholes," Plouffe said on ABC. "And I think the country would be well-served by tax and entitlement reform, because it'll help our economy."

Beyond the economy and the budget, Plouffe indicated that two social issues will be a focus at the outset of the president's second term: immigration and gun control.

On gun control, he mixed statements of optimism with an acknowledgement of political realities. Republicans control the House, and even some Democrats in the Senate have been extremely cautious in addressing the issue.

"It's going to be very, very hard," Plouffe said on CBS's "Face the Nation."

Republican Sen. John Barrasso of Wyoming underscored that point. He said he doubted supporters could get 60 votes in the Senate for legislation allowing universal background checks for gun purchasers and for limiting gun magazines to 10 rounds and under.

"The debt and spending. That's where people are focused. That's the big anxiety of this country," Barrasso said on CNN.

Sen. Roy Blunt, R-Mo., would not answer whether he could support background checks for every gun purchase. Without getting into specifics, he advocated for better information-sharing to prevent some people with mental health problems from buying guns.

"Let's do things that will make a difference here, rather than take one more opportunity to go at an old agenda," Blunt, a gun-rights advocate, said on Fox News Sunday.

When it comes to overhauling the nation's immigration laws, Plouffe said he believes there's broader support from Republicans nationally than there is from Republicans in Congress. Still, "the stars are aligned" for a bill to include beefing up border security as well as giving those already in the U.S. illegally a path to citizenship. He cited business organizations and religious leaders as key players backing a comprehensive immigration bill.

In leading up to Monday's inauguration, the White House has sought to leave the impression that the president will seek common ground with Republicans. It will also seek to undertake public campaigns to build pressure on Congress to act on the administration's legislative priorities.

The new mobilizing effort will be led by a tax-exempt group called Organizing for Action. The move represents the first time a sitting president has ever transformed his presidential campaign operation into an outside group with the express purpose of promoting his agenda. "We're also going to bring the American people more into the debate than we did in the first term," Plouffe said.

Blunt said the president should put forward proposals that a divided Congress can support rather than put in place an ongoing campaign effort designed to pressure Republicans into going along with his priorities.

"I don't think there's any reason to believe from looking at the last four years that that produces much of a result," he said. "It might produce a stalemate. It might get the president re-elected, but he's not running for a third term."

Four tires blow as commuter jet lands

Authorities say a United Express plane from western New York blew four tires as it landed at Newark Liberty International Airport and veered off a runway.

Port Authority of New York and New Jersey spokesman Ron Marsico says Flight 4480 from Rochester was landing Sunday night in New Jersey when several rear tires blew. He tells the Star-Ledger that the plane veered onto a taxiway and didn't strike anything.

The plane was carrying eight passengers and five crew members. No one was hurt.

More news from NBCNewYork.com

It wasn't the only mishap at the Newark airport on Sunday.

Earlier in the day, a United Airlines employee became pinned between a luggage cart and a food service truck. Marsico tells The Record that the worker was seriously hurt. It's unclear how the accident happened.

Quadruple DNA helix discovered in human cells

Sixty years after James Watson and Francis Crick established that DNA forms a double helix, a quadruple-stranded DNA helix has turned up.

Quadruple helices that intertwine four, rather than two, DNA strands had been made in the laboratory, but were regarded as curiosities as there was no evidence that they existed in nature. Now, they have been identified in a range of human cancer cells.

The four-stranded packages of DNA, dubbed G-quadruplexes, are formed by the interaction of four guanine bases that together form a square. They appear to be transitory structures, and were most abundant when cells were poised to divide. They appeared in the core of chromosomes and also in telomeres, the caps on the tips of chromosomes that protect them from damage.

Because cancer cells divide so rapidly, and often have defects in their telomeres, the quadruple helix might be a feature unique to cancer cells. If so, any treatments that target them will not harm healthy cells.

"I hope our discovery challenges the dogma that we really understand DNA structure because Watson and Crick solved it in 1953," says Shankar Balasubramanian of the University of Cambridge, UK.

Tagged with antibodies

Balasubramanian's team identified the four-stranded structures in cancer cells with the help of an antibody that attaches exclusively to G-quadruplexes. To stop them from unravelling into the ordinary DNA, they exposed the cells to pyridostatin, a molecule that traps quadruple helices wherever they form.

This enabled the researchers to count how many formed at each stage of cell multiplication. The G-quadruplexes were most abundant in the "S-phase" ? when cells replicate their DNA just prior to dividing.

"I expect they will also exist in normal cells, but I predict that there will be differences with cancer cells," says Balasubramanian. His hunch is that the G-quadruplexes are triggered into action by chaotic genomic mutations and reorganisations typical of cancerous or precancerous cells.

"This research further highlights the potential for exploiting these unusual DNA structures to beat cancer, and the next part of this is to figure out how to target them in tumour cells," says Julie Sharp of Cancer Research UK, which funded the research.

Another important question that Balasubramanian's and other teams will try to answer is whether G-quadruplexes play a role in embryo development, and whether such a role is mistakenly reactivated in cancer cells. "We plan to find out whether the quaduplexes are a natural nuisance, or there by design," he says.

HDHomeRun Prime update to DLNA starts beta

Got your HDHomeRun Prime, your favorite DLNA client and an itch to watch some premium HD cable TV? Well, then it's time to head over to the beta forum, download the latest firmware then register as a tester. Those of you lucky enough to have a nice TV provider that doesn't lock everything down with Copy Once flag will be able to watch live TV on just about any DLNA playback device that supports MPEG2. Those not so lucky will need a PS3 or a 2012 Samsung HDTV, without which this little gem ain't going to do you much good. Sadly, this doesn't work on the regular HDHomeRun because of hardware limitations, but the new tuners due this spring will allow DLNA access of ATSC, QAM and DVB sources -- as well as allow playback on Media Players that only support h.264. The usefulness of this is still pretty limited while we wait for the rest of SiliconDust's Project:Connect to come together, but we don't see any reason why you wouldn't join in on the fun.
